香菇及猴頭菇β-葡聚糖的制備及其理化性質的研究

摘要:為探索β-葡聚糖的提取方法,本實驗在借鑒已有β-葡聚糖分離技術的基礎上,采用酶法輔助熱水浸提方法從香菇和猴頭菇中提β-葡聚糖,優化β-葡聚糖提取條件,并對其理化性質和結構進行測定。對β-葡聚糖的提取純化工藝改進,以期提高產品收率、降低成本。首先對香菇及猴頭菇使用75%乙醇75℃滅內源酶處理,同時除去一些小分子可溶性糖類;接著對固定進行酶解處理,提取菌菇中的β-葡聚糖,與此同時,考察酶解時間、酶解溫度、酶解pH、加酶量以及三種酶(纖維素酶、果膠酶和木瓜蛋白酶)的復配比例對β-葡聚糖提取率的影響。采用苯酚硫酸法測定多糖含量,利用多糖含量和β-葡聚糖得率設計正交試驗,通過正交試驗的所得結果,得出猴頭菇β-葡聚糖提取的最優方案:三種酶的復配比例為1:1:1,加酶量為1.5%,酶解pH值為5,酶解溫度為65℃,酶解時間為60min;香菇β-葡聚糖提取的最優方案:三種酶的復配比例為1:1:1,加酶量為2.0%,酶解pH值為5,酶解溫度為55℃,酶解時間為70min。高效液相過濾色譜法測定相對分子質量,從香菇和猴頭菇相對分子量分布圖可看出,猴頭菇β-葡聚糖的純度比香菇β-葡聚糖純度相對較高;紅外光譜法測定其結構,根據紅外光譜分析可得知猴頭菇和香菇中通過酶法輔助熱水浸提提取出來的多糖是以β-葡聚糖為主的雜多糖。35086

Study on Preparation of mushrooms and Hericium beta glucan and its physicochemical properties

High performance liquid phase filtration chromatography determination of relative molecular mass, from Lentinus edodes and Hericium erinaceus relative molecular weight distribution diagram can be seen, hedgehog hydnum mushroom beta glucan purity than the mushroom and the purity of the beta glucan is relatively high; infrared spectrometry determination of the structure, based on the infrared spectrum analysis can learn that Hericium erinaceus and Lentinus edodes by enzyme assisted hot water extraction to extract polysaccharide is based on beta glucan polysaccharide.
