菌株SQR9野生型和突變體在不同IAA合成中間產物吲哚類物質中產IAA能力的探究

摘要:解淀粉芽袍桿菌(Bacillus amyloliquefaciens)SQR9是本實驗室從黃瓜根際分離的一株促生菌株,對黃瓜專化型病原菌尖袍鐮刀菌有很強的拮杭功能。在盆栽和大田試驗中,該菌株表現出良好的促生和土傳病害防控效果,其與腐熟堆肥制成的微生物有機肥在農業生產中已得到廣泛應用。生物依賴色氨酸合成IAA的幾種途徑中,都有其特定的中間產物,根據這些特定中間產物分別命名為吲哚丙酮酸途徑、色胺途徑、吲哚乙酰胺途徑、吲哚乙腈途徑途徑。本實驗通過在解淀粉芽袍桿菌SQR9和突變體ΔysnE中添加各種途徑的中間產物,分析SQR9和ΔysnE的發酵液的IAA和這7種關鍵組分,從而尋找基因ysnE參與IAA合成的途徑。經過實驗分析得出結論:吲哚丙酮酸(IPA)途徑和色胺(TAM)途徑可能是SQR9依賴色氨酸合成IAA最主要的兩個途徑。基因ysnE可能分別參與IAA合成途徑中的吲哚丙酮酸途徑和色胺途徑,而且是在吲哚丙酮酸到吲哚乙醛之間及色胺到吲哚乙醛之間發揮功能。基因ysnE沒有參與吲哚乙酰胺(IAM)途徑和吲哚乙腈(IAN)途徑。35417

Abstract:Bacillus amyloliquefaciens SQR9, isolated from the rhizosphere of cucumber, can control Fusarium wilt of cucumber. The strain was proved to be an outstanding plant growth-promoting rhizobacteria (PGPR) strain by its ability to inhibit the growth of Fusarium oxysporum f. sp. cucumerinum J. H. Owen (FOC) and decrease the incidence of Fusarium wilt in field experiments, and a bioorganic fertilizer made of organic fertilizer fermented with B. amyloliquefaciens SQR9 is widely used in agricultural production.Biological dependence on tryptophan synthesis of IAA in several ways, has its specific intermediate, according to the specific intermediate respectively named IPA, TAM, IAM,and IAN.This experiment through the solution of starch bud robe coli SQR9 and mutant ΔysnE, various ways of intermediate product is added to the analysis of SQR9 and ΔysnE fermented liquid of IAA and the seven key components, the ysnE to search for genes involved in IAA synthetic way. The conclusion of the experiment is:IPA and TAM approach may be the two main pathways of the SQR9 dependence on tryptophan synthesis IAA. Gene ysnE may participate in IAA respectively synthetic ways of indole pyruvate way and tryptamine way, and it is between indole pyruvate and indole acetaldehyde and serotonin function between indole acetaldehyde. Gene ysnE is not involved in IAM and IAN.
