摘要:本文用化學還原法制備RE微粒、納米銀微粒,研究了納米銀標記二抗的pH、二抗標記量、NC膜的預處理、一抗的濃度、封閉劑的種類、銀染液濃度、RE與Ag的混合比例對銀染效果的影響,進行了檢出限的測定。采用紫外-可見分光光度計(UV-Vis)對納米銀、Ag-二抗、RE微粒、RE-Ag、RE-Ag-二抗進行表征。用Zeta電位儀分析了納米銀的電位。結果表明:硝酸纖維素膜進過預處理后,銀染效果好;納米銀標記二抗的合適pH為7.4;加入二抗最少量5µg時,Ag-二抗復合物穩定存在;在一抗濃度為5μg/mL、銀染液濃度為1.82×10-3 mol/L硝酸銀和18.2×10-3 mol/L對苯二酚等體積混合時,銀染效果較好。La 、Sm 、Ce、 Nd 用于與納米銀混合形成RE-Ag-二抗,銀染效果好;在RE:Ag(RE:La、Ce、Sm、Nd)分別為4:6、3:7、2:8、1:9時,灰度值較大,其中La-Ag微粒的銀染效果最好,檢出限最低,為0.1fg/mL。本文創新之處:用稀土取代部分納米銀提高試劑盒的靈敏度。35855

For LA, SM, CE, Nd and nano silver mixture formation RE-Ag- second antibody, silver staining effect is good; in RE:Ag (re: La, CE, SM, nd) were 4:6, 3:7, 2:8 And 1:9, gray value is larger, the La-Ag particles of the silver dyeing effect is best, the minimum detection limit for 0.1fg/mL.
